A phase I trial, an early-stage human study designed to test safety and look for signs of effect, gave white button mushroom powder to 36 men with recurrent prostate cancer. Why Aromatase Matters for Estrogen Dominance in Menopause

Estrogen dominance is common through perimenopause and beyond, and the reason comes down to ratios. Progesterone production falls earlier and faster than estrogen as ovulation becomes irregular, which leaves estrogen with less opposition even when its absolute levels are declining. Symptoms like heavy or erratic cycles, breast tenderness, poor sleep, irritability, and stubborn weight gain around the middle often show up during this window.

Once the ovaries slow their output, the body's estrogen production shifts to other tissues, and fat tissue running the aromatase conversion becomes the main source of circulating estrogen.[1] More fat tissue means more aromatase activity, more aromatase activity means more estrogen made outside the ovaries, and that estrogen goes unopposed because progesterone is low. The original City of Hope researchers pointed at exactly this, noting that a mushroom-rich diet could moderate aromatase activity and reduce the estrogen being made directly in tissue in postmenopausal women.[1]

Agaritine: Why I Never Eat Button Mushrooms Raw

Raw button mushrooms contain agaritine, a naturally occurring nitrogen compound in the hydrazine family that the mushroom produces on its own. Fresh samples have tested at roughly 90 to 200 milligrams per pound, with the highest concentrations in the cap skin and gills.[5] Harm in humans from eating raw button mushrooms has not been demonstrated, and the mushroom has never been restricted as a food. Understanding what agaritine does once it enters the body explains why I cook it out anyway.

What Agaritine Does in the Body

The concern with agaritine is what your body converts it into after you swallow it. An enzyme in your kidneys called gamma-glutamyl transpeptidase clips off part of the agaritine molecule, and the piece left behind is a free hydrazine, a small reactive nitrogen compound named 4-(hydroxymethyl)phenylhydrazine. Enzymes in the liver then oxidize that hydrazine into something more reactive still, a diazonium ion, which toxicology researchers identify as the end point of this chain.[7]

That ion falls apart on contact with living tissue, and as it does it releases a free radical, an unstable fragment that reacts with whatever it touches. In lab studies those fragments break DNA strands and attach themselves to guanine, one of the four building blocks of DNA, forming damage sites that chemists call DNA adducts.[7],[8] Damage of this kind is what can lead to mutations, and agaritine's breakdown products cause mutations in standard lab tests using bacteria, which is why researchers treat the hydrazine family with caution [in vitro evidence].[7]

The animal evidence is mixed in a way worth spelling out. Early mouse studies feeding raw Agaricus mushrooms or agaritine-related hydrazines reported increased tumor development, while a later feeding study in rats and mice looked for the predicted DNA damage sites after actual agaritine ingestion and did not find them, and detected no mutation-causing activity in the animals' urine [animal evidence].[8] The open question is how efficiently the human body runs this conversion chain at food doses. Nobody has answered that, and I would rather not volunteer for the experiment when an hour in a pot settles it.

Every step of that conversion chain requires intact agaritine reaching your digestive tract, so heat that breaks the molecule apart in the pot leaves your enzymes nothing to work with. Agaritine degrades in the refrigerator, in the freezer, and fastest of all with cooking. A Czech research team measured this across household methods and found that frying in butter or oil cut agaritine by 35 to 70 percent, while five minutes of boiling pulled about half of it into the cooking liquid and destroyed another quarter outright. After roughly two hours of continued cooking, only about 10 percent of the original agaritine remained.[5] Commercial canning drops it more than tenfold.[6]

That research is the logic behind my one hour minimum. Some agaritine degrades from heat directly, and some leaches into the liquid the mushrooms release as they cook. In a pot method like mine that liquid stays in the pan, simmers, and reduces, so the leached agaritine keeps breaking down under continued heat. A quick five minute sauté leaves a meaningful share of the compound behind, while an hour of low, steady cooking takes you most of the way to the two hour numbers and concentrates the flavor as it goes.

Do the Hormone-Supportive Compounds Survive the Cooking?

A fair question at this point is whether an hour of heat destroys the compounds you are cooking the mushrooms for in the first place, and whether the pot needs to be drained. The answer to both is no. Aromatase never enters the pot at all, since it is an enzyme your own body produces in fat tissue, the ovaries, and other sites. What the mushroom contributes are the inhibitor compounds, mainly conjugated linoleic acid and related unsaturated fatty acids.[2]

Those fatty acids hold up well under cooking temperatures. The original screening research specifically prepared heat-stable extracts, meaning the aromatase-inhibiting activity survived heat processing by design.[1] Draining would move you in the wrong direction, since fatty acids concentrate in the butter and the reduced cooking liquid. Serve the mushrooms in their glaze and you keep the full package: the agaritine cooked down, the fatty acids retained.

Slow-Cooked Garlic Butter Mushrooms

I make this with two 24 oz containers of white button mushrooms from Costco, which comes to ~3 lbs. That sounds like a lot until you watch them cook down to a fraction of their starting volume. This keeps in the refrigerator for days and goes with eggs, steak, sourdough, or straight out of the pot. Mushrooms are cultivated in controlled conditions on pasturized compost, and white mushrooms are on EWG's clean 15 list, so I don't really worry about buying organic.

Slow-Cooked Garlic Butter Mushrooms

Ingredients

Instructions

  1. Melt the butter in a large heavy pot over medium heat.
  2. Add all the mushrooms at once. You can chop them first or leave them whole and break them apart with your spoon as they soften. Whole mushrooms hold a better texture through the long cook.
  3. Stir to coat the mushrooms in butter. Within 15 minutes they will release a large amount of liquid. Keep it in the pot, since the aromatase-inhibiting fatty acids concentrate there and the liquid will reduce back down as it simmers.
  4. Cook uncovered for 1 hour minimum at a steady simmer, stirring every 10 to 15 minutes and lowering the heat if anything begins to stick.
  5. In the last 15 minutes, once the liquid has mostly reduced, stir in the garlic, thyme, rosemary, oregano, salt, and pepper. Adding the garlic late keeps it from turning bitter over the long cook.
  6. The mushrooms are done when they are deep brown, dense, and coated in a glossy butter glaze rather than sitting in liquid. Do not drain. Serve them glaze and all.

Store leftovers in a sealed glass container in the refrigerator for up to 5 days. They reheat well in a dry skillet.
